</p><p><b>Lactase</b> oder <b>Laktase</b> (<a href="Gen" title="Gen">Gen</a>-Name <i>LCT</i>, (englisch) auch <i>Lactase-<a href="Phlorizin" title="Phlorizin">Phlorizin</a>-<a href="Hydrolase" class="mw-redirect" title="Hydrolase">Hydrolase</a></i>, Abkürzung <i>LPH</i>) heißt ein im <a href="Darm" title="Darm">Darm</a> vorkommendes <a href="Enzym" title="Enzym">Enzym</a>, das <a href="Lactose" title="Lactose">Lactose</a> (Milchzucker) in ihre Bestandteile <a href="Galactose" title="Galactose">Galactose</a> (Schleimzucker) und <a href="Glucose" title="Glucose">Glucose</a> (Traubenzucker) spaltet. Ohne diese chemische Reaktion können die Bestandteile des Milchzuckers nicht durch die Dünndarmschleimhaut aufgenommen werden. Beim Menschen wird das Enzym normalerweise im Kindesalter im <a href="D%C3%BCnndarm" title="Dünndarm">Dünndarm</a> produziert, in Europa bei den meisten Menschen auch später im Erwachsenenalter. Ein Mangel an Lactase kann mehrere Ursachen haben (siehe <a href="Laktoseintoleranz" title="Laktoseintoleranz">Laktoseintoleranz</a>) und führt bei zwei Drittel der betroffenen Personen zu Verdauungsproblemen, wenn Milchzucker aufgenommen wird.
</p><p>Die Lebensmittelindustrie verwendet Enzyme, die ebenfalls Lactose hydrolysieren können und dem Oberbegriff <a href="%CE%92-Galactosidase" title="Β-Galactosidase">β-Galactosidasen</a> zugeordnet werden. Im wissenschaftlichen Sprachgebrauch sollte Lactase nur für β-Galactosidasen aus Säugetieren verwendet werden.
Da die Herstellung menschlicher Lactase unwirtschaftlich wäre, werden ausschließlich mikrobielle β-Galactosidasen aus sicheren und zugelassenen Bakterien (<i>Escherichia coli</i> K-12), Hefen (<i>Kluyveromyces lactis</i>) oder filamentösen Pilzen biotechnisch für die Lebensmitteltechnik hergestellt.<sup id="cite_ref-2" class="reference"><a href="#cite_note-2"><span class="cite-bracket">[</span>2<span class="cite-bracket">]</span></a></sup> Die <a href="Hydrolyse" title="Hydrolyse">Hydrolyse</a> von Lactose führt zur Reduktion des Milchzuckers, wobei Glucose und Galactose entstehen, die in lactosefreier Milch zu einer erhöhten Süße beitragen. Milchzuckerfreie Lebensmittel werden nicht nur an lactoseintolerante Personen verkauft, milchzuckerfreies Eis hat beispielsweise günstigere Verarbeitungseigenschaften und durch andere Kristallisationseigenschaften eine feinere Beschaffenheit.<sup id="cite_ref-3" class="reference"><a href="#cite_note-3"><span class="cite-bracket">[</span>3<span class="cite-bracket">]</span></a></sup><sup id="cite_ref-Born_1997_4-0" class="reference"><a href="#cite_note-Born_1997-4"><span class="cite-bracket">[</span>4<span class="cite-bracket">]</span></a></sup>
</p><p>Neben der Lactaseaktivität hat das Enzym eine weitere <i>Glycosylceramidase</i>-Aktivität, weshalb es in der Fachliteratur als Lactase-Phlorizinhydrolase bezeichnet wird. Strukturell besteht das <i>LCT</i>-Gen aus vier ähnlichen Domänen, wobei die Lactaseaktivität an der vierten und die Phlorizinhydrolase an der dritten Domäne stattfindet.<sup id="cite_ref-5" class="reference"><a href="#cite_note-5"><span class="cite-bracket">[</span>5<span class="cite-bracket">]</span></a></sup>
</p><p>Als integrales <a href="Membranprotein" title="Membranprotein">Membranprotein</a> ist LPH in der <a href="B%C3%BCrstensaum" class="mw-redirect" title="Bürstensaum">Bürstensaummembran</a> der säulenförmig angeordneten <a href="Hauptzelle" class="mw-redirect" title="Hauptzelle">Hauptzellen</a> des Zottenepithels des Dünndarms aller Säugetiere lokalisiert. <a href="Mutation" title="Mutation">Mutationen</a> im <i>LCT</i>-Gen können über eine Verringerung der LPH-Aktivität Ursache für erbliche Varianten von <a href="Lactoseintoleranz" class="mw-redirect" title="Lactoseintoleranz">Lactoseintoleranz</a> sein. Gewisse andere Mutationen führen dagegen über eine Erhöhung der LPH-Produktion bzw. ihrer Beibehaltung im Erwachsenenalter zur <b>Laktasepersistenz</b> vor allem bei Bewohnern der nördlichen Hemisphäre. Der Normalzustand jedoch ist eine Verringerung der LPH-Produktion nach der <a href="Stillzeit" class="mw-redirect" title="Stillzeit">Stillzeit</a>.
</p>
<div class="mw-heading mw-heading2"><h2 id="Evolution">Evolution</h2></div>
<p>Aufgrund der vierfachen Glycosidase-1-Domäne ist von zwei <a href="Genverdopplung" class="mw-redirect" title="Genverdopplung">Genverdopplungen</a> in der Evolution des <i>LCT</i>-Gens auszugehen. Dass die letzte dieser Verdopplungen noch vor der Entwicklung der <a href="Wirbeltiere" title="Wirbeltiere">Wirbeltiere</a> stattfand, ist der Tatsache zu entnehmen, dass bereits die zu <i>LCT</i> orthologen Gene in mehreren Fischarten vier Glycosidase-1-Domänen aufweisen.<sup id="cite_ref-6" class="reference"><a href="#cite_note-6"><span class="cite-bracket">[</span>6<span class="cite-bracket">]</span></a></sup>
</p><p>Üblicherweise wird bei Menschen Lactase nur in Säuglingen gebildet. In Nordeuropa wird Lactase auch in 90 % der Erwachsenen gebildet.<sup id="cite_ref-spektrum-765816_7-0" class="reference"><a href="#cite_note-spektrum-765816-7"><span class="cite-bracket">[</span>7<span class="cite-bracket">]</span></a></sup> Für die Evolution der <i>Lactasepersistenz</i>-Mutation, die zur Herstellung von Lactase auch in Erwachsenen führt, ist ihre <a href="Selektion_(Evolution)" title="Selektion (Evolution)">Selektion</a> in milchtrinkenden Kulturen ursächlich. Die von Beja-Pereira und anderen propagierte <i>Gen-Kultur-Koevolution</i> ist heute die wahrscheinlichste Erklärung für den hohen Anteil der Persistenz in der menschlichen Bevölkerung und ihr Verbreitungsprofil.<sup id="cite_ref-PMID14634648_8-0" class="reference"><a href="#cite_note-PMID14634648-8"><span class="cite-bracket">[</span>8<span class="cite-bracket">]</span></a></sup> Die Laktasepersistenz-Mutation breitete sich in Nordeuropa innerhalb von 3000 Jahren von etwa 10 % der Erwachsenen auf 90 % der Erwachsenen aus.<sup id="cite_ref-spektrum-765816_7-1" class="reference"><a href="#cite_note-spektrum-765816-7"><span class="cite-bracket">[</span>7<span class="cite-bracket">]</span></a></sup>

<p>Das für LPH <a href="Genetischer_Code" title="Genetischer Code">codierende</a> <a href="Gen" title="Gen">Gen</a> <i>LCT</i> liegt auf dem <a href="Chromosom_2_(Mensch)" title="Chromosom 2 (Mensch)">zweiten Chromosom</a> (2q21). Das <i>LCT</i>-Gen erstreckt sich über 17 <a href="Exon" title="Exon">Exons</a> und 49.340 Basenpaare. Nach der <a href="Transkription_(Biologie)" title="Transkription (Biologie)">Transkription</a> entsteht <a href="MRNA" title="MRNA">mRNA</a>, die 6274 Basen enthält und zu einem 1927 <a href="Aminos%C3%A4uren" title="Aminosäuren">Aminosäuren</a> langen Molekül Pre-pro-LPH <a href="Translation_(Biologie)" title="Translation (Biologie)">übersetzt</a> wird. Dieses enthält vier ähnliche Glycosidase-1-<a href="Proteindom%C3%A4ne" title="Proteindomäne">Domänen</a> und eine <a href="Signalsequenz" title="Signalsequenz">Signalsequenz</a>, die bewirkt, dass Prepro-LPH vom ER zur <a href="Zellmembran" title="Zellmembran">Zellmembran</a> transportiert wird. <p>Lactose wird in Glucose und Galactose <a href="Hydrolyse" title="Hydrolyse">gespalten</a>. Die optimalen Bedingungen für die menschliche Laktase liegen bei einem <a href="PH-Wert" title="PH-Wert">pH-Wert</a> von sechs<sup id="cite_ref-pmid6786877_13-0" class="reference"><a href="#cite_note-pmid6786877-13"><span class="cite-bracket">[</span>13<span class="cite-bracket">]</span></a></sup> und einer Temperatur von 37 °C.<sup id="cite_ref-pmid17512743_14-0" class="reference"><a href="#cite_note-pmid17512743-14"><span class="cite-bracket">[</span>14<span class="cite-bracket">]</span></a></sup>

<div class="mw-heading mw-heading2"><h2 id="Industrielle_Nutzung">Industrielle Nutzung</h2></div>
<p>Kommerziell produzierte Lactase wird aus <a href="Hefen" title="Hefen">Hefepilzen</a> wie <i><a href="Kluyveromyces_fragilis" class="mw-redirect" title="Kluyveromyces fragilis">Kluyveromyces fragilis</a></i> gewonnen. Das so gewonnene Enzym wird in Form von Tabletten und Kapseln angeboten, damit Menschen mit Lactoseintoleranz Milchprodukte zu sich nehmen können. Auch gibt es lactosefreie Milch, bei welcher die Lactose bereits durch Zusatz von Lactase aufgespalten wurde; damit ist diese Milch für Menschen genießbar, denen das Enzym Lactase fehlt.
</p><p>In der Herstellung von <a href="Speiseeis" title="Speiseeis">Speiseeis</a> wird Lactase eingesetzt, da die Spaltprodukte Glucose und Galactose süßer sind als die Lactose und somit der Zuckerzusatz eingeschränkt werden kann. Außerdem kristallisiert Lactose bei Temperaturen, wie sie im Speiseeis vorliegen, während die Spaltprodukte weiterhin gelöst vorliegen und dem Eis so eine feinere Beschaffenheit verleihen.
</p><p>Mengenangaben werden in <a href="Food_Chemicals_Codex" title="Food Chemicals Codex">FCC</a>-Einheiten ausgedrückt. 1000 FCC-Einheiten können unter optimalen Bedingungen 5 Gramm Milchzucker abbauen.<sup id="cite_ref-15" class="reference"><a href="#cite_note-15"><span class="cite-bracket">[</span>15<span class="cite-bracket">]</span></a></sup> Dabei kommt es unter anderem auf die Viskosität der Lösung an. In dickflüssigen Lösungen wird eine größere Menge des Enzyms benötigt.
Da Milch im sauren Milieu des Magens ausflockt, benötigt man ebenfalls eine größere Menge Lactase, um die Lactose vollständig aufzuspalten, wenn man Lactase direkt beim Konsum von Milchprodukten zu sich nimmt.
